What the gene does
CYP1A2 builds the enzyme that demethylates caffeine in the liver. It handles the overwhelming majority of the job, which is unusual: most compounds are cleared by several routes at once. Here one gene does nearly all of it, which is why a single variant has such a visible effect.
What the variant changes
rs762551 sits in the first intron and changes how strongly the gene is induced. A/A carriers clear caffeine quickly. C-carriers hold onto it, so the same cup circulates longer and at a higher effective concentration.
The research here is unusually clean for nutrigenetics. In a case-control study, slow metabolisers under 59 drinking four or more cups a day had more than double the odds of myocardial infarction, while fast metabolisers showed no such association and in some analyses appeared protected. A separate cohort found nearly threefold higher hypertension incidence in slow metabolisers drinking more than three cups a day.
What it means for you
If you're a slow metaboliser, the useful move is a ceiling and a cut-off time rather than giving up coffee. Afternoon caffeine is the part that tends to matter, both for blood pressure and for sleep. It's also worth counting the hidden sources: tea, cola, energy drinks and some painkillers.
If you're fast, you have more room, though the ceiling still exists and it's personal.
