What the gene does
Reactive oxygen species are a normal by-product of using oxygen. Around 5% of the oxygen your cells take up ends up as superoxide or a related species. Clearing it takes a relay: SOD2 converts superoxide into hydrogen peroxide, then GPX1 converts that into water using glutathione as the reducing partner.
GPX1 is a selenoprotein. Selenium isn't a cofactor it can do without: the amino acid selenocysteine sits in the enzyme's active site.
What the variant changes
Pro198Leu is a missense change that alters how responsive the enzyme is to selenium supply. A randomised controlled trial found the rise in glutathione peroxidase activity after selenium supplementation depended on genotype, so the same intake produced different enzyme activity in different people.
The pattern repeats across cohorts in New Zealand, the United States and Brazil, including a Brazil nut supplementation study. Genotype effects are most visible where baseline selenium is lower, which is much of Europe.
What it means for you
Selenium has a narrower useful range than most micronutrients: too little limits your antioxidant enzymes, too much is genuinely toxic. That's why the answer is a measured dose rather than a large one.
Diet does a lot of the work. Brazil nuts are the densest source by a wide margin, and fish and eggs contribute steadily. Glutathione support matters alongside it, since GPX1 spends glutathione to do its job. N-acetylcysteine keeps that recycling system topped up.
What your formula does
Your selenium dose is set from this genotype together with your oxidative stress chapter, since both draw on the same defence chain. It stays within safe limits regardless of genotype.
